    The influence of survivin shRNA on the cell cycle and the invasion of SW480 cells of colorectal carcinoma

    <p>Abstract</p> <p>Background</p> <p>The objective was to understand the influence of Survivin plasmid with short hairpin RNA (shRNA) on the cell cycle, invasion, and the silencing effect of Survivin gene in the SW480 cell of colorectal carcinoma.</p> <p>Methods</p> <p>A eukaryotic expression vector, PGCH1/Survivin shRNA, a segment sequence of Survivin as target, was created and transfected into colorectal carcinoma cell line SW480 by the non-lipid method. The influence on the Survivin protein was analyzed by Western blotting, while the cell cycle, cell apoptosis were analyzed by flow cytometry, and invasion of the cell was analyzed by Transwell's chamber method.</p> <p>Results</p> <p>After the transfection of PGCH1/Survivin shRNA, the expression of Survivin protein in SW480 cells was dramatically decreased by 60.68%, in which the cells were stopped at G2/M phase, even though no apoptosis was detected. The number of transmembranous cells of the experimental group, negative control group, and blank control group were 14.46 ± 2.11, 25.12 ± 8.37, and 25.86 ± 7.45, respectively (P <<it>0.05</it>).</p> <p>Conclusion</p> <p>Survivin shRNA could significantly reduce the expression of Survivin protein and invasion of SW480 cells. Changes in cell cycle were observed, but no apoptosis was induced.</p

    Extracellular Vesicles: “Stealth Transport Aircrafts” for Drugs

    Extracellular vesicles (EVs) can deliver many types of drugs with their natural source material transport properties, inherent long-term blood circulation capabilities and excellent biocompatibility, and have great potential in the field of drug carrier. Modification of the content and surface of EVs according to the purpose of treatment has become a research focus to improve the drug load and the targeting of EVs. EVs can maximize the stability of the drugs, prevent immune clearance and achieve accurate delivery. Therefore, EVs can be described as “stealth transport aircrafts” for drugs. This chapter will respectively introduce the application of natural EVs as cell substitutes in cell therapy and engineered EVs as carriers of nucleic acids, proteins, small molecule drugs and therapeutic viral particles in disease treatment. It will also explain the drug loading and modification strategies of EVs, the source and characteristics of EVs. In addition, the commercialization progress of EVs drugs will be mentioned here, and the problems in their applications will be discussed in conjunction with the application of EVs in the treatment of COVID-19

    Intelligent Information Dissemination Scheme for Urban Vehicular Ad Hoc Networks

    In vehicular ad hoc networks (VANETs), a hotspot, such as a parking lot, is an information source and will receive inquiries from many vehicles for seeking any possible free parking space. According to the routing protocols in literature, each of the vehicles needs to flood its route discovery (RD) packets to discover a route to the hotspot before sending inquiring packets to the parking lot. As a result, the VANET nearby an urban area or city center may incur the problem of broadcast storm due to so many flooding RD packets during rush hours. To avoid the broadcast storm problem, this paper presents a hotspot-enabled routing-tree based data forwarding method, called the intelligent information dissemination scheme (IID). Our method can let the hotspot automatically decide when to build the routing-tree for proactive information transmissions under the condition that the number of vehicle routing discoveries during a given period exceeds a certain threshold which is calculated through our developed analytical packet delivery model. The routing information will be dynamically maintained by vehicles located at each intersection near the hotspot if the maintaining cost is less than that of allowing vehicles to discover routes themselves. Simulation results show that this method can minimize routing delays for vehicles with lower packets delivery overheads

    Comparative study on appropriate drought and flood index selection in a tropical farming island in China

    The traditional drought and flood analysis method had not fully considered the proportion analysis of different drought and flood grades in the historical years of each rainfall station. This made results unconvincing and made it difficult to deeply understand the characteristics and applicability of various methods. Based on the daily rainfall data of 88 stations in Hainan Island from 1970 to 2019, the China-Z index and the Standardized Precipitation Index (SPI) were used to compare and analyze the spatial and temporal distribution characteristics of droughts and floods from three different time scales (flood season, non-flood season and the whole year). The results showed that both SPI and China-Z index can well reflect the actual drought and flood situations in Hainan Island. The analysis of the proportions of different drought and flood grades in the historical years of each rainfall station and regional historical drought and flood statistics suggested that the China-Z index had a better indication effect than SPI on the extreme drought and flood grades. The alternation of drought and flood between different eras were obvious. Hainan Island generally presented an east-west reverse drought-flood variation trend, as well as a north-south reverse drought-flood variation trend. The drought and flood in the central mountainous area of Hainan Island had been relatively stable. The distribution pattern of drought and flood had a good spatial consistency in the three periods. On the whole, Hainan Island had shown a trend of flood in the east and drought in the west in the past 50 years

    Superconductivity in the vicinity of antiferromagnetic order in CrAs

    One of the common features of unconventional, magnetically mediated superconductivity as found in the heavy-fermions, high-transition-temperature (high-Tc) cuprates, and iron pnictides superconductors is that the superconductivity emerges in the vicinity of long-range antiferromagnetically ordered state.[1] In addition to doping charge carriers, the application of external physical pressure has been taken as an effective and clean approach to induce the unconventional superconductivity near a magnetic quantum critical point (QCP).[2,3] Superconductivity has been observed in a majority of 3d transition-metal compounds,[4-9] except for the Cr- and Mn-based compounds in the sense that the low-lying states near Fermi level are dominated by their 3d electrons. Herein, we report on the discovery of superconductivity on the verge of antiferromagnetic order in CrAs via the application of external high pressure. Bulk superconductivity with Tc ~ 2 K emerges at the critical pressure Pc ~ 8 kbar, where the first-order antiferromagnetic transition at TN = 265 K under ambient pressure is completely suppressed. Abnormal normal-state properties associated with a magnetic QCP have been observed nearby Pc. The close proximity of superconductivity to an antiferromagnetic order suggests an unconventional pairing mechanism for the superconducting state of CrAs. The present finding opens a new avenue for searching novel superconductors in the Cr and other transitional-metal based systems
